The PhIFI Core Facility at the Brody School of Medicine/ECU houses a fully equipped Amersham. GE Healthcare Typhoon 9410 Imager for 2D imaging of phosphorescent, chemiliumscent and fluorescent gel and array data.

Typhoon 9410

 

The Typhoon 9410 is capable of detecting, imaging and quantifying five different forms of emission: phosphor autoradiography, chemiluminescence, blue-excited fluorescence, green-excited fluorescence, and red-excited fluorescence. Thus, in addition to radioactivity detection like the old Phosphorimager, it allows direct fluorescence and phosphorescence measurement. All imaging modes have a linear dose response of five orders of magnitude. It can also scan to a resolution of 10 um, making the Typhoon 9410 unique in its ability to image microarrays.

Examples of Routine Typhoon Imager Uses:

  • Radioactivity detection from a blot or dried  gel (32P, 33P, 35S, 14C, etc.; requires Phosphorimage Screen)

 

  • Chemiluminescent/Fluorescent detection on western blots using ECL+

 

  • Multiple fluorophore labeled proteins, individual detection (e.g. Cy3 vs Cy5)

 

  • Fluorescent tag labeled DNA detection (e.g. ROX, TAMRA, etc.)

 

  • Imaging of slide-mounted DNA or Protein Microarrays

 

  • Protein detection (in gel) by Sypro Ruby stain
