The purpose of these core facilities is to provide a service to east and west campus research faculty for a minimal fee for service.

 

Flow Cytometry/Confocal Microscopy Facility:

Purpose:  To provide multiple data acquisition and analysis capabilities in flow cytometry, confocal microscopy, and laser scanning cytometry.  To train faculty, staff and students in operation of these instruments and assist in computer data analysis and interpretation.

 

Capabilities: 

 

Flow Cytometry

  • Immunophenotyping
  • Cell cycle analysis
  • Up to five-color fluorescence analysis
  • Clonal sorting and deposition
  • Calcium and magnesium intracellular flux
  • Membrane potential studies

Confocal Microscopy

  • Localization and co-localization of biomolecules in tissues or cells
  • 3-D reconstruction of fluorescently labeled cells or tissues

Laser Scanning Cytometry

Similar capabilities as flow cytometry (except sorting) on a slide or

multiwell plate format, with up to four-color fluorescence analysis


Laser Capture Microdissection- Zeiss PALM LMPC

Instrument which allows excision and removal of specific target cells from a tissue
or live cell culture

 

Instrumentation:

  • Polychromatic Flow Cytometry System (Four Laser, Ten Color)-
    Becton Dickinson LSR II
  • Three bench-top flow analyzers (three color) - Becton Dickinson FACScan
  • Cell sorter - Becton Dickinson FACSVantage SE with high-speed sorting
    cabability (Turbo-sort), with three lasers, extending from UV to far
    red wavelengths
  • Computer workstations with zip and CD-RW data storage
  • Confocal Microscope- Zeiss LSM 510 with 3 lasers and 3 fluorescence
    detectors
  • Laser Scanning Cytometer- CompuCyte iCys (location- Brody 7S-19)
  • Laser Capture Microdissection System- Zeiss PALM LMPC
    (location- Brody 3S-30)

 

Electron Microscopy Laboratory:

Location:  7W39 BrodyMedicalSciencesBuilding

Purpose:  To provide research support in transmission electron microscopy (TEM)  

 

Capabilities:  

  • Training in the use of the TEM   
  • Training in routine sample preparation and staining 

 

Instrumentation:  

  • JEOL 1200EX TEM; CCD camera [Fees for EM scope time]
  • Leica UC6 ultramicrotome  [Training at no cost until 09/2007]

 

Histology Laboratory:

Purpose:  To provide research support in histology and immunohistochemistry

 

Capabilities:

  • Training in sectioning and staining

 

Fee for service: 

Embedding and sectioning (frozen and paraffin), histology staining, immunohistochemistry, and knife sharpening

  • Microm HM505E Cryostat
  • Microm HM340E digital advance rotary microtome
  • Citadel 1000 Tissue Processor for paraffin embedding
  • Sequenza Immunostaining System

Metabolomics Core Facility:

PhIFI Core Facility (Phosphor-Imaging/Fluorescence Imaging):

Instrumentation: 

  • Amersham/ GE Healthcare Typhoon 9410 Imager
  • Dell GX270 dedicated to scanner workstation

 

Capabilities:

  • Radioactivity detection from a blot or dried gel (32P,33P, 35S, 14C, etc.; requires Phosphorimage screen)
  • Chemiluminescent/Fluorescent detection on western blots using ECL+
  • Multiple fluorophore labeled  proteins, individual detection (e.g. Cy3 vs Cy5)
  • Fluorescent tag labeled DNA detection (e.g. ROX, TAMRA, etc.) on wet gels between plates
  • Imaging of slide-mounted DNA or Protein microarrays
  • Protein detection (in gel) by Sypro Ruby stain
